Significance of Measurement: Nitrite reductase (NiR) is widely distributed in microorganisms and plants and is a key enzyme in the nitrogen cycle. It reduces nitrite to NO or NH₃, thereby decreasing nitrite accumulation and associated toxicity.
Principle of Measurement: NiR reduces NO₂^- to NO, resulting in a decrease in NO₂^- available for diazotization reactions that produce a purple-red compound. Changes in absorbance at 540 nm reflect NiR activity.
