Paneth cells are located at the base of intestinal glands and are morphologically similar to intestinal epithelial cells. They are pyramidal in shape, typically arranged in small clusters, with oval nuclei positioned basally. The cytoplasm contains coarse eosinophilic granules, resulting in red staining in hematoxylin–eosin preparations. Paneth cell granules contain zinc, arginine, and tyrosine. Immunocytochemical studies have demonstrated the presence of lysozyme within these granules, suggesting a role in bacterial regulation within the intestinal lumen, although the full functional profile of Paneth cells remains incompletely defined.
This staining kit employs the Lendrum method. Phloxine B stains Paneth cells red, while Mayer’s hematoxylin counterstains nuclei blue. The staining is stable, produces high contrast, and is suitable for paraffin-embedded tissue sections.
