Papanicolaou staining is a routine cytological method originally developed for assessing estrogenic status in vaginal epithelium and detecting genital tract pathogens. Combined use of Orange G6 with EA36 or EA50 produces distinct green, blue, and pink cytoplasmic staining.
The EA36-based Papanicolaou staining solution uses EA36 for cytoplasmic staining and a non-toxic modified hematoxylin for nuclear staining. Compared with EA50, EA36 is more suitable for gynecological cytology specimens, including cervical cancer and precancerous lesion screening. Nuclei appear blue to black, while keratinized squamous cell cytoplasm appears pink to orange-red.
