Glycogen staining is a routine technique in histopathology. The periodic acid–Schiff (PAS) reaction was first introduced by McManus in 1946 for the visualization of mucins and is widely used to detect glycogen and other polysaccharides. In addition to glycogen, PAS staining highlights neutral mucosubstances, certain acidic substances, cartilage, pituitary tissue, fungi, molds, pigments, amyloid, and basement membranes.
Periodic acid oxidizes adjacent 1,2-glycol groups in carbohydrates to form aldehydes, which subsequently react with Schiff reagent to produce an insoluble magenta-colored compound. PAS stain is primarily applied for histological detection of glycogen and polysaccharides.
