PAS staining is a routine pathological method for detecting glycogen and other polysaccharides. Periodic acid oxidizes vicinal glycol groups to aldehydes, which react with Schiff reagent to form a magenta product. The technique also highlights neutral mucins, basement membranes, fungi, cartilage, pituitary tissue, and amyloid. Oxidation conditions must be carefully controlled to avoid overoxidation. PAS staining provides high sensitivity, strong specificity, and relatively rapid processing.
