Peroxidases are heme-containing oxidoreductases located mainly in peroxisomes. Using hydrogen peroxide as an electron acceptor, they catalyze oxidation of phenolic and amine substrates.
The DAB-based peroxidase staining method, recommended by the International Council for Standardization in Haematology (ICSH), relies on enzymatic oxidation of 3,3′-diaminobenzidine (DAB), resulting in a brown insoluble precipitate at sites of peroxidase activity. This method is applicable to blood, bone marrow, and cytological smears.
