Anthocyanins are water-soluble natural pigments belonging to the flavonoid family and are widely distributed in plant roots, stems, leaves, flowers, and fruits, imparting red to purple coloration.
This assay uses the pH differential method. At pH 1.0, anthocyanins exhibit maximal absorbance at 530 nm, whereas at pH 4.5 they convert to colorless chalcone forms. Absorbance differences at 530 nm and 700 nm under different pH conditions enable accurate quantification while minimizing interference.
