Nitrate nitrogen is the primary nitrogen source for plants. The nitrate nitrogen content in plant tissues reflects the availability of nitrate nitrogen in soil and serves as an indicator of soil nitrogen fertilization status. Determination of nitrate nitrogen in plants provides important information on nitrogen nutritional status and is also relevant for evaluating the quality of vegetables and plant-derived food products.
Principle: Under strongly acidic conditions, nitrate ions (NO3-) react with salicylic acid to form nitrosalicylic acid. In alkaline conditions (pH > 12), the product exhibits a yellow color, the intensity of which is proportional to nitrate concentration within a defined range. Nitrate nitrogen content is determined by colorimetric measurement.
