Superoxide anions reduce nitroblue tetrazolium (NBT) to an insoluble blue formazan, allowing localization of reactive oxygen species.
This method is suitable for fresh plant tissues such as root tips and leaves, where superoxide-rich regions appear blue to dark blue.
