Significance of Measurement: Polygalacturonase (EC 3.2.1.15) is a cell wall-associated enzyme that hydrolyzes α-(1,4)-polygalacturonic acid in pectin, leading to cell wall disassembly and fruit softening. It is involved in fruit ripening, leaf and flower abscission, pathogen defense, cell expansion, and lignification, and is of high research value in plant disease resistance and food preservation.
Principle of Measurement: Polygalacturonase hydrolyzes pectic acid to produce galacturonic acid with reducing aldehyde groups. These react with DNS reagent to form a red-brown product with a characteristic absorbance at 540 nm. Enzyme activity is calculated from absorbance changes at 540 nm.
