Significance of Measurement: Proline is widely present in animals, plants, microorganisms, and cultured cells. Under stress conditions, proline accumulates significantly in plants and serves as an indicator of stress tolerance, particularly drought resistance.
Principle of Measurement: Proline is extracted using sulfosalicylic acid and reacts with acidic ninhydrin upon heating to form a red product. After toluene extraction, absorbance is measured at 520 nm.
