Principle of Measurement: Under strong alkaline conditions, peptide bonds react with Cu²⁺ to form a purple complex. The color intensity is proportional to protein concentration and independent of molecular weight or amino acid composition. The assay range is 1–10 mg protein, suitable for samples with high protein concentrations.
