Hemosiderin is an iron-containing pigment derived from hemoglobin degradation, appearing as golden-yellow to brown granules. Perls’ Prussian blue reaction detects ferric iron by forming an insoluble blue ferric ferrocyanide complex following treatment with potassium ferrocyanide and dilute acid.
This method specifically distinguishes hemosiderin from other pigments and is commonly used to assess hemorrhagic lesions and iron deposition in tissues. The staining solution is stable, suitable for long-term storage, and compatible with counterstaining. Staining intensity depends on ferric iron content.
