Prussian blue staining detects non-heme iron deposits by forming blue ferric ferrocyanide precipitates, commonly used for bone marrow, liver, spleen, and kidney tissues. The method distinguishes hemosiderin from other pigments but has limited sensitivity for very low iron concentrations. Nuclear fast red is used as a classic counterstain to enhance morphological detail.
