Pyruvate carboxylase (PC, EC 6.4.1.1) is present in mitochondria of animals, fungi, and yeast. It catalyzes the conversion of pyruvate, ATP, CO2, and water to oxaloacetate, ADP, and Pi and is the first rate-limiting enzyme in gluconeogenesis.
PC-generated oxaloacetate is converted to malate by malate dehydrogenase with concomitant oxidation of NADH to NAD+. The decrease in absorbance at 340 nm reflects PC activity.
