Significance of Measurement: Pyruvate dehydrogenase (PDH, EC 1.2.4.1) is the rate-limiting enzyme of the pyruvate dehydrogenase complex and links glycolysis to the tricarboxylic acid cycle.
Principle of Measurement: PDH catalyzes pyruvate oxidation while reducing 2,6-dichlorophenolindophenol (DCPIP), resulting in decreased absorbance at 600 nm. PDH activity is determined by monitoring this decrease.
