Significance of Measurement: Pyruvate kinase (PK, EC 2.7.1.40) catalyzes the final step of glycolysis and is one of the major rate-limiting enzymes responsible for ATP generation.
Principle of Measurement: PK catalyzes the conversion of phosphoenolpyruvate and ADP to pyruvate and ATP. Lactate dehydrogenase further catalyzes the reduction of pyruvate to lactate, accompanied by NADH oxidation. The decrease in absorbance at 340 nm reflects PK activity.
