Reduced glutathione (GSH) is the major intracellular thiol antioxidant and plays critical roles in redox regulation, thiol protection, and amino acid transport. The GSH/GSSG ratio is a key indicator of cellular redox status.
DTNB reacts with GSH to form a chromophoric product with absorbance at 412 nm. Absorbance is proportional to GSH concentration. Measurement of GSH, GSSG, and their ratio reflects intracellular redox balance.
