Reducing sugars are widely present in animals, plants, microorganisms, and cultured cells. In plants, reducing sugars mainly include glucose, fructose, and maltose, which are the most common monosaccharides and disaccharides. Among them, glucose and fructose are not only the primary substrates for respiration but also serve as precursors for the further synthesis of sucrose, starch, and cellulose.
Upon heating, reducing sugars react with 3,5-dinitrosalicylic acid (DNS) in an alkaline solution to form a brown-red amino compound with a characteristic absorption peak at 540 nm. Within a certain concentration range, the reducing sugar content shows a linear relationship with the absorbance at 540 nm. Based on the standard curve, the reducing sugar content in the sample can be quantitatively determined.
