This method is based on the adsorption of ammoniacal silver by tissue proteins, followed by formaldehyde reduction to metallic silver deposited on reticular fibers. Unlike traditional protocols, this modified method omits gold toning and sodium thiosulfate treatment, resulting in enhanced contrast.
The procedure includes oxidation, bleaching, mordanting, silver impregnation, and reduction. It is widely used for differentiating tumor origin and type, including carcinoma versus sarcoma, lymphosarcoma versus reticulum cell sarcoma, vascular tumors, Ewing sarcoma, meningioma, astrocytoma, malignant peripheral nerve sheath tumors, and early infiltrative carcinoma.
