This method detects cellular senescence based on increased SA-β-Gal activity. Using X-gal as a substrate, senescent cells produce an insoluble blue precipitate visible under light microscopy. The assay is applicable to cultured cells and tissue sections and selectively stains senescent cells without labeling presenescent, quiescent, immortalized, or tumor cells.
