Significance: Soluble starch synthase (SSS, EC 2.4.1.21) is typically present in the plastid stroma in a soluble form and catalyzes starch chain elongation, primarily contributing to amylopectin synthesis.
Principle: SSS catalyzes the transfer of glucose from ADP-glucose (ADPG) to a starch primer (glucan), producing ADP. In the presence of pyruvate kinase, hexokinase, and glucose-6-phosphate dehydrogenase, NADP+ is sequentially reduced to NADPH. The amount of NADPH formed is proportional to ADP production. SSS activity is calculated by measuring the increase in NADPH absorbance at 340 nm.
