Significance: Starch branching enzyme (SBE, EC 2.4.1.18) is a key enzyme involved in amylopectin biosynthesis in plants. Measurement of SBE activity is important for starch biosynthesis studies, crop quality improvement, and genetic breeding research.
Principle: Starch forms a complex with iodine that exhibits characteristic absorbance at 660 nm. SBE cleaves branch points in amylopectin, reducing the absorbance of the starch–iodine complex. The percentage decrease in absorbance over a defined time reflects SBE activity.
