Significance: Starch is the primary storage carbohydrate in plants. Its quantification is important for evaluating food nutritional value and investigating carbohydrate metabolism in plant tissues.
Principle: Soluble sugars are removed using 80% ethanol, separating them from starch. Starch is then hydrolyzed into glucose under acidic conditions. Glucose content is determined by the anthrone colorimetric method, allowing calculation of starch concentration.
