Significance of Measurement: Succinate dehydrogenase (SDH, EC 1.3.5.1) is a mitochondrial marker enzyme located on the inner mitochondrial membrane. It serves as a critical link between the electron transport chain and oxidative phosphorylation and also provides electrons for respiratory chains in prokaryotes.
Principle of Measurement: SDH catalyzes the oxidation of succinate to fumarate. The released hydrogen is transferred via phenazine methosulfate (PMS) to reduce 2,6-dichlorophenolindophenol (DCPIP), which exhibits a characteristic absorption at 600 nm. SDH activity is determined by monitoring absorbance changes at 600 nm.
