Lipids include neutral fats, lipoids, and their derivatives. They are insoluble in water but readily soluble in organic solvents such as ethanol and ether. Lipids in the human body include storage lipids (e.g., neutral fats) and structural lipids (e.g., phospholipids, glycolipids, cholesterol). Neutral fats consist of glycerol esterified with three fatty acids.
Sudan dyes (Sudan II, III, IV, Sudan Black B) and Oil Red O are commonly used for lipid staining. Sudan Black B is a lipid-soluble dye that dissolves into lipid-containing structures in the cytoplasm, staining lipids, phospholipids, and cholesterol brown-black to dark black.
After staining, neural phospholipids and brain phospholipids show positive reactions, whereas renal clear cell carcinoma and ovarian fibroma typically show negative reactions.
