Acid phosphatase is a lysosomal marker enzyme with optimal activity at pH 4.5–5.5. TRAP activity is particularly associated with osteoclasts. Using naphthol AS-BI phosphate as substrate, ACP activity produces a colored azo precipitate localized in the cytoplasm. TRAP-positive cells retain staining in the presence of tartrate. This method is applicable to blood smears, cell preparations, and tissue sections.
