Significance: Sugars are essential plant components and primary substrates and storage forms for metabolism. Total sugars include soluble mono- and disaccharides, as well as insoluble starch, cellulose, and chitin.
Principle: Acid hydrolysis releases reducing sugars, which react with DNS in NaOH and glycerol under heat to form orange-red amino compounds. Absorbance at 540 nm reflects total sugar content.
