Significance: Serum total cholesterol represents the total cholesterol content within all lipoproteins and includes free cholesterol and cholesterol esters. The liver is the primary organ responsible for cholesterol synthesis and storage. Cholesterol is essential for steroid hormone synthesis, bile acid production, vitamin D formation, and cell membrane structure. Serum TC concentration serves as an indicator of lipid metabolism.
Principle: Cholesterol esters are hydrolyzed by cholesterol esterase to free cholesterol and fatty acids. Free cholesterol is oxidized by cholesterol oxidase to generate cholestenone and hydrogen peroxide. In the presence of peroxidase, hydrogen peroxide reacts with 4-aminoantipyrine and phenol to form a red quinone compound. Color intensity is proportional to TC concentration.
