Significance: Trehalase (THL, EC 3.2.1.28) is widely present in animals, plants, microorganisms, and cultured cells. Its primary function is to hydrolyze trehalose into glucose, which is directly used for energy supply.
Principle: The 3,5-dinitrosalicylic acid (DNS) method is used to measure reducing sugars produced from trehalose hydrolysis catalyzed by THL. Reducing sugars react with DNS upon heating to form a brown-red amino compound. Within a certain range, color intensity is proportional to reducing sugar concentration, reflecting THL activity.
