Triglycerides are lipid molecules composed of long-chain fatty acids and glycerol and serve as major membrane components and respiratory substrates. TG is extracted with isopropanol and saponified by KOH to release glycerol. Glycerol is oxidized by periodate to formaldehyde, which reacts with acetylacetone in the presence of chloride ions to form a yellow compound with absorbance at 420 nm. Color intensity correlates with TG content.
