Significance: UDP-glucose pyrophosphorylase (UGP) is a key enzyme in glycogen and polysaccharide biosynthesis. It catalyzes the activation of glucose by converting glucose-1-phosphate and UTP into UDP-glucose, an essential precursor for glycogen synthesis.
Principle: UGP catalyzes a reversible reaction generating glucose-1-phosphate. In a coupled enzymatic system involving phosphoglucomutase and glucose-6-phosphate dehydrogenase, NADP+ is reduced to NADPH. The rate of increase in absorbance at 340 nm reflects UGP activity.
