Fc receptor (FCRN) isan approximately 45 kDa transmembrane glycoprotein with structural homology toMHC class I proteins. It is widely expressed in endothelial and epithelial cellsand plays an important role in IgG homeostasis. It does not bind the structurallyrelated chicken IgY. FCRN additionally binds to albumin, and both it and IgG arebound at pH 5.0 but not at pH 8.0. FCRN associates noncovalently with beta 2Microglobulin,and this interaction is important for the intracellular trafficking of FCRN. FCRNcycles between the plasma membrane and acidified intracellular compartments ofendothelial cells and epithelial cells. It binds endocytosed IgG and albumin inthe low pH vesicles and transports them to the plasma membrane for extracellularrelease at higher Ph. FCRN is also expressed in neutrophils and myeloid antigenpresenting cells. It can enhance IgGmediated phagocytosis and antigen presentationby these cells, but it promotes the degradation of opsonizing IgG rather thanreturning it to the circulation. |
