Description : | This gene is a member of the immunoglobulin superfamily and encodes a protein which transmits an inhibitory signal to T cells. The protein contains a V domain, a transmembrane domain, and a cytoplasmic tail. Alternate transcriptional splice variants, encoding different isoforms, have been characterized. The membrane-bound isoform functions as a homodimer interconnected by a disulfide bond, while the soluble isoform functions as a monomer. Mutations in this gene have been associated with insulin-dependent diabetes mellitus, Graves disease, Hashimoto thyroiditis, celiac disease, systemic lupus erythematosus, thyroid-associated orbitopathy, and other autoimmune diseases. |

Q&As (5)
Ask a questionYes, several clinical trials are investigating the combination of CTLA-4 inhibitors with PD-1 inhibitors and other immunotherapies to improve the overall effectiveness of cancer treatment.
CTLA-4 is expressed on various immune cells, including T cells, regulatory T cells (Tregs), and some B cells, contributing to its diverse immunoregulatory functions.
Modulating CTLA-4 may have applications in organ transplantation by suppressing the immune response against transplanted tissues and reducing the risk of rejection.
CTLA-4 is involved in the suppression of autoreactive T cells, contributing to the prevention of autoimmune responses by maintaining immune tolerance.
While both CTLA-4 and PD-1/PD-L1 inhibitors enhance anti-tumor immune responses, they target different checkpoints in the immune system. CTLA-4 primarily regulates T-cell activation in the early stages, whereas PD-1/PD-L1 acts later in the immune response.
