Description : This gene is a member of the R-spondin family and encodes a secreted activator protein with two cystein-rich, furin-like domains and one thrombospondin type 1 domain. In mice, the protein induces the rapid onset of crypt cell proliferation and increases intestinal epithelial healing, providing a protective effect against chemotherapy-induced adverse effects.
Source : HEK293
Species : Human
Tag : His
Form : Lyophilized from 0.22 μm filtered solution in PBS, pH7.4. Normally Trehalose are added as protectants before lyophilization.
Molecular Mass : Biotinylated RSPO1, fused with 6×His tag at the C-terminus, has a calculated MW of 27.6 kDa. The predicted N-terminus is Ser 21 . DTT-reduced Protein migrates as 40 kDa due to glycosylation.
Endotoxin : Less than 1.0 EU per μg of the Biotinylated RSPO1 by the LAL method.
Purity : >95% as determined by reduced SDS-PAGE.
Storage : Avoid repeated freeze-thaw cycles. No activity loss was observed after storage at: In lyophilized state for 1 year (4°C); After reconstitution under sterile conditions for 3 months (-70°C).

Ask a question
What distinguishes RSPO1 from other RSPO molecules, particularly in terms of its potential therapeutic use in Regenerative Medicine? 05/31/2022

RSPO1 stands out for its potential therapeutic use in Regenerative Medicine due to its mitogenic activity in stem cells.

What methods were employed to characterize the purified rhRSPO1, both structurally and biologically? 02/11/2022

Characterization methods included Western blotting, mass spectrometry, and in vitro (C2C12 cells) and in vivo (BALB/c mice) biological activity assays, confirming structural integrity and biological efficacy.

What tissue-specific processes are regulated by RSPOs, and what are some examples of these processes? 08/25/2021

RSPOs regulate various tissue-specific processes, including bone formation, skeletal muscle tissue development, proliferation of pancreatic β-cells, intestinal stem cells, and cancer progression.

Describe the purification strategy used to obtain rhRSPO1, including the chromatographic steps involved. 07/08/2021

The purification strategy for rhRSPO1 involved two sequential chromatographic steps: heparin affinity chromatography followed by molecular exclusion chromatography.

How does RSPO1 induce the Wnt pathway, and what are the downstream effects on cell proliferation, differentiation, and death? 05/19/2021

RSPO1 induces the Wnt pathway by stabilizing the Wnt receptor complex, leading to enhanced Wnt signaling and influencing cellular processes.

How was recombinant human RSPO1 (rhRSPO1) generated, and which cell line was used for this purpose? 04/08/2018

rhRSPO1 was generated using the HEK293 cell line, and the coding sequence was synthesized and subcloned into a mammalian cell expression vector.

What is the main function of R-Spondin proteins, including RSPO1, in cellular processes? 04/14/2017

R-Spondin proteins, including RSPO1, play crucial roles in cell proliferation, differentiation, and death by inducing the Wnt pathway.
