About Common gamma Chain Receptor Family Ligands

Common gamma chain (γc) is a transmembrane protein encoded by the CD132 gene. It is a common signal transducer for many cytokine receptors, including IL-2, IL-4, IL-7, IL-9, IL-15, and IL-21, etc. γc has two isoforms: long-chain γc and short-chain γc. Long-chain γc is found in T cells, B cells, and natural killer (NK) cells, while short-chain γc is found mainly in monocytes and dendritic cells. Both isoforms are structurally similar, consisting of a single transmembrane protein with an extracellular domain that binds to ligands and an intracellular region involved in signaling.

γc plays a key role in cytokine signaling. Together with cytokine-specific receptors, it forms the corresponding cytokine receptor complex, which activates the signaling pathway. The signaling pathways activated by this family include the PI3-K-Akt pathway, the Ras-MAPK pathway, and the Jak- STAT pathway. γc deletion leads to X-linked severe combined immunodeficiency (X-SCID), a disease in which the immune system is severely impaired and susceptible to infection and death. The discovery of common γ chain receptors provides a new therapeutic approach to treat X-SCID by using cytokines with long-chain γc. For example, cytokines such as IL-2, IL-4, and IL-7, together with γc, can promote the proliferation and differentiation of T, B, and NK cells, thus restoring immune function. In addition, for other immune diseases and tumors, γc can also be used to develop new therapeutic approaches.
