Overview
This gene encodes a member of the vascular endothelial growth factor receptor (VEGFR) family. VEGFR family members are receptor tyrosine kinases (RTKs) which contain an extracellular ligand-binding region with seven immunoglobulin (Ig)-like domains, a transmembrane segment, and a tyrosine kinase (TK) domain within the cytoplasmic domain. This protein binds to VEGFR-A, VEGFR-B and placental growth factor and plays an important role in angiogenesis and vasculogenesis. Expression of this receptor is found in vascular endothelial cells, placental trophoblast cells and peripheral blood monocytes. Multiple transcript variants encoding different isoforms have been found for this gene. Isoforms include a full-length transmembrane receptor isoform and shortened, soluble isoforms. The soluble isoforms are associated with the onset of pre-eclampsia.[provided by RefSeq, May 2009] -

What is flt1 protein?
FLT1 stands for FMS-like tyrosine kinase 1, also known as vascular endothelial growth factor receptor 1 (VEGFR1). It is a type I transmembrane receptor tyrosine kinase that binds VEGF-A, VEGF-B, and PlGF ligands with high affinity. FLT1 plays a key role in blood vessel formation (vasculogenesis) and maturation (angiogenesis) during embryonic development and tissue remodeling.
What is the function of flt1 protein?
Angiogenesis: FLT1 signaling promotes endothelial cell proliferation, migration and survival, key steps in sprouting angiogenesis and blood vessel branch formation.
Vasculogenesis: During embryonic development, FLT1 helps in de novo blood vessel formation through vasculogenesis, the process of assembling endothelial progenitor cells into primitive vessels.
Vascular permeability: FLT1 activation increases permeability of endothelial cell junctions, allowing plasma components and cells to exit vessels during angiogenesis.
Flt1 related signaling pathway
PI3K-Akt pathway: FLT1 dimerization leads to recruitment and phosphorylation of PI3K, which activates PDK1 and Akt. Akt promotes cell survival, growth and metabolism.
PLCγ pathway: PLCγ is activated by FLT1 and generates IP3 and DAG. IP3 causes calcium release while DAG activates PKC. This regulates cell migration, survival and permeability.
Ras-Raf-MEK-ERK pathway: FLT1 activates GRB2/SOS complex which activates Ras. This triggers the Raf-MEK-ERK cascade, promoting cell proliferation and survival.
SRC family kinases: FLT1 recruits and activates c-SRC and other SRC kinases. They induce downstream signaling regulating migration, sprouting and permeability.
Focal adhesion kinase (FAK) pathway: FLT1 induces FAK phosphorylation which triggers reorganization of actin cytoskeleton and focal adhesions, controlling cell motility.
