Description:  The acetylation of histone lysine residues plays a crucial role in the epigenetic regulation of gene transcription. Acetylated lysine residues are recognized by a small protein domain known as a bromodomain. Plant Homeodomain (PHD) fingers are zinc binding domains comprised of ~ 50 amino acids and have a Cys4-His-Cys3 motif. PHD fingers are found in more than 100 nuclear proteins that play a role in regulating chromatin. The C-terminal PHD and bromodomain regions of SP140 work in concert to bind to chromatin and ultimately regulate gene transcription. Nuclear bodies, which are involved in the pathogenesis of acute promyelocytic leukemia and viral infection, consist of several components, such as Sp100, NDP52, PML, and SP140. SP140 may play a role in the pathway that inactivates HIV-1 lacking viral infectivity factor (VIF).
