What Is mRNA Sequencing?

mRNA sequencing (mRNA-Seq) is a targeted RNA-Seq protocol that enriches for all polyadenylated (poly-A) transcripts of the transcriptome. Only around 1~2% of the entire transcriptome is comprised of poly-A tailed RNA, which is the coding part of the genome. mRNA-Seq enables the identification of novel transcripts, mRNA selective splicing, and gene fusion events. This technique also provides another affordable approach for gene expression quantification and differential gene expression analysis between sample groups. By targeting mRNA, sequencing depth is improved as resources are dedicated to the sequencing of coding genes. This makes identifying rare variants and low expressed mRNA transcripts easier. With high throughput and high sensitivity, it is a powerful tool for in-depth study of transcripts. At present, mRNA-Seq has been widely utilized in gene screening of animals and plants, as well as disease research. In mRNA-Seq, single RNA strands are converted to complementary DNAs (cDNA) and then turned into double stranded DNA before being sequenced. So, while the initial starting input material is RNA, material loaded on the sequencing instrument is DNA.
