Global DNA methylation analysis is often utilized as a screening tool prior to gene-specific or in-depth assays. Various methods for assessing DNA methylation at a global scale include pyrosequencing, end-specific PCR, high performance liquid chromatography (HPLC), liquid chromatography-tandem mass spectrometry (LC-MS/MS), LUminometric methylation assay (LUMA), M.SssI acceptance assay, and global DNA methylation 5mC quantification ELISA. The assay based on enzyme-linked immunosorbent assay (ELISA) enables rapid assessment of global DNA methylation status. ELISA-based assays are typically prone to high variability; thus, they are only suitable for the rough estimation of DNA methylation. Still, they are quick and easy to perform methods that serve well for the identification of large changes in global DNA methylation.

What Is Global DNA 5mC ELISA Quantification?

Immunoassay based on microtitration plates is known as ELISA and is a mature method that can be effectively applied to detect epigenetic modifications of DNA immobilized on plastics, using highly specific antibodies to target epigenetic markers. Briefly, the genomic DNA is denatured, fixed, and the methylated cytosines are detected through sequential incubations steps with: a) a primary antibody raised against 5mC; b) a labeled secondary antibody; and then c) colorimetric/fluorometric detection reagents.
