Description:  Recombination hotspots are small segments of the genome where genetic information gets shuffled at an elevated rate during meiosis. PR domain zinc finger protein 9 or PRDM9 is a histone methyltransferase that binds specifically to these hotspots catalyzing trimethylation of H3K4 in nucleosomes near its binding site, thus initiating genetic recombination by recruiting recombination initiation machinery. PRDM9 contains an amino terminal PR/SET domain for HTMase activity and a C-terminal tandem array of twelve C2-H2 zinc-fingers. The zinc fingers of PRDM9 are thought to bind with high affinity to a 13-mer hotspot motif in DNA, which promotes double strand break formation in the area around the binding site. PRDM9 is unique in that its activity is tissue specific, only expressing in germ cells during meiosis.
