Scientific Background:  Jumonji lysine demethylases (JMJDs) are oxygenases catalyzing demethylation of mono-, di-, or trimethylated lysines, and its members (JMJD2A, JMJD2B, JMJD2C, and JMJD2D) are Fe(II)- and alpha-ketoglutarate-dependent enzymes harboring a JmjC catalytic domain. JMJD2A (JHDM3 or KDM4A) catalyzes the demethylation of di- and trimethylated H3K9 and H3K36 and participate in transcriptional repression of ASCL2 and E2F-responsive promoters via the recruitment of HDACs and NCOR1, respectively. JMJD2A as well as JMJD2C have been suggested to act as co-activators of AR-dependent gene transcription. JMJD2A is widely expressed in human tissues and cell lines with high expression in human T-cell lymphotropic virus 1-infected cell lines, HT1376 bladder carcinoma cell line, U2OS osteosarcoma cell line and prostate cancer cell lines. JMJD2A is overexpressed in mouse and human lung cancers has been proposed as an oncogene that represents a target for Ras-expressing tumors. Silencing of JMJD2A gene has been shown to result in cell cycle change and proliferation inhibition, and suppression of tumor cell migration/ invasion.
