Description:  U-937 nuclear extract (TPA stimulated) was prepared from cell cultures of the human histiotic lymphoma U-937 cell line. The U-937 cell line was derived from malignant cells of a pleural effusion of a 37-year old male patient. These cells are of myeloid lineage and differentiate into a mature macrophage phenotype in response to various stimuli. U-937 cells produce and secrete large quantities of cytokines and chemokines either spontaneously or in response to treatment with soluble stimuli, such as TNFα. In addition, IFNβ and IFNϒ stimulation has been shown to induce the expression of chemokine receptors on the cell surface of U-937 cells and results in increased production of interleukins. Because U-937 cells are one of only a few human cell lines with characteristic monocytic properties, they are commonly used in research as in vitro models for monocyte/macrophage differentiation. Treatment of U-937 cells with the potent tumor-promoting phorbol ester 12-O-tetradecanoylphorbol-13-acetate (TPA) leads to various biological changes that mimic those observed in cells transformed by chemical carcinogens or viruses. In mice, TPA has been shown to induce long lasting effects, including inflammation and hyperplasia, that are associated with the critical first step in multistage tumorigenesis known as tumor conversion. Thus, TPA stimulated U-937 cells are frequently utilized in cancer research as a model system for studying the radiomimetic and tumor-promoting activity of TPA on replicating cells.

Extract Composition:  U-937 nuclear extract (TPA stimulated) is supplied in Dilution Buffer (20 mM Hepes (pH 7.9), 100 mM KCl, 1 mM MgCl2, 20% glycerol, 0.5 mM PMSF and 0.5 mM DTT). Cells were cultured in medium supplemented with 5 nM TPA (phorbol, 12-myristate, 13 acetate) for 72 hours at 37°C immediately prior to harvesting.
Application Notes:  U-937 extract is recommended for studies of monocyte/macrophage differentiation. U-937 nuclear extract (TPA stimulated) is specifically recommended for analysis of the mechanisms underlying tumorigenesis or TPA-mediated tumor conversion and its associated biochemical responses, including increased membrane metabolism, induction of protein biosynthesis, and alterations in the cell cycle.
Storage:  Store at -80°C. Aliquot to avoid repeated freezing and thawing.
