Description:  HeLa nuclear extract (TNF-α stimulated) was prepared from a cell culture of the HeLa human epithelial carcinoma cell line. Stimulation of HeLa cells with the tumor necrosis factor-alpha (TNF-α), a member of a group of cytokines that stimulate the acute phase reaction of the immune response, leads to programmed cell death, or apoptosis, and induction of activation of the NFκB signal transduction pathway. The transcription factor NFκB is widely studied due to its implication in the regulation of genes that control inflammation, cell proliferation and cell survival.

Components:  2 x 100 µg of HeLa nuclear extract (TNF-α stimulated) at 2.5 µg/µl.
Extract Composition:  HeLa nuclear extract was collected in Lysis Buffer after a 30-minute incubation with TNF-α (20 ng/ml). The Lysis Buffer consists of 20 mM Hepes pH 7.5, 350 mM NaCl, 20% glycerol, 1% Igepal-CA630, 1 mM MgCl2, 0.5 mM EDTA and 0.1 mM EGTA, 5 mM DTT and protease inhibitors. The protein content has been determined by a Bradford-based assay.
Application Notes:  HeLa nuclear extract (TNF-α stimulated) is specifically recommended for analysis of apoptosis and the role of NFκB activation in 1) inflammation, 2) cell growth, and 3) cell survival.
Storage:  Store at -80°C. Aliquot to avoid repeated freezing and thawing.
