ChIP-qPCR combines chromatin immunoprecipitation (ChIP) technology with real-time quantitative PCR to study protein-DNA interactions at known genomic binding sites. ChIP technology utilizes antibody specificity to enrich DNA fragments bound to specific transcription factor/histone modification; qPCR technology uses SYBR fluorescent dye, non-specifically incorporating into DNA double strands, emitting fluorescence, to ensure that the increase of fluorescence signal and increased PCR products are completely synchronized. And the finally quantitative analysis is carried out using Ct value. Because of the lowest cost of performing qPCR, ChIP-qPCR is an attractive strategy, which is advantageous for the research of interrogating specific genes and potential regulatory regions in a wide range under different experimental conditions.
