• Description: T-cell surface glycoprotein CD3 epsilon chain, also known as CD3e, T3e, TCRe and IMD18, is a single-pass type I transmembrane protein. CD3E contains 1 Ig-like (immunoglobulin-like) domain in the extracellular region and 1 cytoplasmic ITAM (immune tyrosine activation motif) domain. CD3 delta chain, also known as CD3d, T3d and IMD19, is a single-pass type I transmembrane protein. CD3d contains 1 Ig-like domain in the extracellular region and 1 cytoplasmic ITAM (immune tyrosine activation motif) domain. CD3e:CD3d is part of the T-cell receptor (TCR)/CD3 complex and is involved in T-cell development and signal transduction. The TCR/CD3 complex of T-lymphocytes consists of either a TCR alpha/beta or TCR gamma/delta heterodimer co-expressed at the cell surface with the invariant subunits of CD3 gamma, delta, epsilon, zeta, and eta. The TCR/CD3 complex plays an important role in coupling antigen recognition to several intracellular signal-transduction pathways leading to the activation of T cells. CD3e plays an essential role in T-cell development. In CD3e-deficient mice, all stages of T cell development and activation that are TCR-dependent were impaired, but not eliminated. Defects in the CD3d gene cause autosomal recessive severe combined immunodeficiency T-cell-negative/B-cell- positive/NK-cell-positive (SCIDBNK) and immunodeficiency 19 (IMD19).
