Case 1: Target-agnostic discovery of membrane receptor targets via phenotypic screening and thermal proteome profiling

This study used a multi-tiered experimental design to identify membrane-bound protein targets of small molecules, focusing on autophagy inhibition. A high-content phenotypic screen in MCF7-eGFP-LC3 cells first identified indophagolin as a potent autophagy inhibitor. Thermal proteome profiling (TPP) and cellular thermal shift assays (CETSA) were employed to identify P2X4 as a membrane-bound target, validated by western blot and isothermal dose-response. Nano-LC-MS/MS with TMT labeling enabled proteomic quantification. Complementary assays, including immunoblotting, confocal microscopy, and calcium flux in P2X/P2Y-expressing cells, confirmed functional inhibition. The workflow demonstrates a target-agnostic approach for de novo identification of membrane receptor targets of bioactive compounds.

Case 2: Rapid protein quantification in oilseeds via modified whole-grain Kjeldahl method

This study presents a modified Kjeldahl method for rapid and efficient protein quantification in oilseed crops, aiming to overcome limitations of traditional sample preparation. Instead of grinding oilseeds into fine flour—a time-consuming step prone to cross-contamination and oil loss—the method directly digests whole grains. A modified digestion mixture was also introduced to reduce frothing and shorten digestion time. The new approach was statistically validated (ANOVA, t-tests, Pearson correlation), showing no significant difference in protein content between whole grain and flour preparations across five oilseed types. This streamlined method enhances speed and reliability in protein analysis.
