Description : Serine/threonine-protein kinase ULK1 involved in autophagy in response to starvation. It acts upstream of phosphatidylinositol 3-kinase PIK3C3 to regulate the formation of autophagophores, the precursors of autophagosomes. It also acts both as a downstream effector and negative regulator of mammalian target of rapamycin complex 1 (mTORC1) via interaction with RPTOR. Activated via phosphorylation by AMPK and also acts as a regulator of AMPK by mediating phosphorylation of AMPK subunits PRKAA1, PRKAB2 and PRKAG1, leading to negatively regulate AMPK activity. Plays a role early in neuronal differentiation and is required for granule cell axon formation.

Ask a question
How does ULK1 interact with other autophagy-related proteins? 03/01/2021

ULK1 interacts with several autophagy-related proteins, including ATG13, FIP200, and ATG101, forming a complex that is essential for the initiation of autophagy.

Are there any known upstream regulators of ULK1 activity? 09/02/2019

mTOR (mechanistic target of rapamycin) is a known upstream regulator of ULK1. When nutrients are abundant, mTOR is active and inhibits ULK1. Conversely, under nutrient starvation, mTOR is inactivated, leading to ULK1 activation and initiation of autophagy.

What is the primary function of the ULK1 protein in cellular processes? 07/27/2019

ULK1 is a serine/threonine-protein kinase that plays a crucial role in the initiation of autophagy, a cellular process responsible for the degradation and recycling of cellular components.

What are the implications of ULK1 dysregulation in diseases like cancer? 07/07/2019

Dysregulation of ULK1 can lead to impaired autophagy, which has been implicated in several diseases, including cancer. Overactive or underactive ULK1 can disrupt cellular homeostasis and contribute to tumorigenesis.

Are there any known post-translational modifications of ULK1 that influence its activity? 01/02/2019

ULK1 undergoes several post-translational modifications, including phosphorylation and ubiquitination, which can either activate or inhibit its kinase activity and, consequently, its role in autophagy.

How does ULK1 play a role in the initiation of autophagy? 09/29/2018

ULK1 is part of a complex that is essential for the initiation of autophagy. Upon activation, ULK1 phosphorylates several targets, leading to the nucleation and formation of the autophagosome.

How does ULK1 activity relate to nutrient availability in cells? 07/06/2018

ULK1 activity is sensitive to nutrient availability. Under nutrient-rich conditions, ULK1 is inhibited, while under nutrient-deprived conditions, ULK1 becomes activated, promoting autophagy to recycle cellular components and generate energy.
