      Case 1: Multiplex bead-based system for biomarker validation

      This study presents a magnetic bead–based multiplex system designed to conserve precious clinical samples while enabling integrated protein and glycan analysis. Antibody-coupled beads were used to capture target proteins, with protein quantification achieved through antibody-based immunoassays and glycan profiling performed via lectin immunosorbent assays (LISAs). Tested on TIMP-1, tissue plasminogen activator, membrane metallo-endopeptidase, and DPP-4, the system matched the performance of conventional single assays while requiring minimal sample input. Applied to prostate cancer tissues, the approach revealed that fucosylated TIMP-1 and DPP-4 better distinguished aggressive from nonaggressive cases, highlighting its utility for biomarker discovery and validation.

      Case 2: Magnetic vs. non-magnetic beads inmalaria serology

      This study compared non-magnetic and magnetic bead–based multiplex assays for measuring antibody responses to five Plasmodium falciparum vaccine candidates. Both bead types successfully coupled recombinant antigens and produced comparable mean fluorescence intensity (MFI) values across tested samples from western Kenya. While stored non-magnetic beads maintained performance, they showed greater aggregation. Magnetic beads, though more costly, offered higher recovery, easier handling, and faster protocol execution. Overall, magnetic beads proved to be a practical and efficient alternative to non-magnetic beads for malaria serology, supporting their use in high-throughput antibody assessment in vaccine studies.
