Overview
The bone morphogenetic proteins (BMPs) are a family of secreted signaling molecules that can induce ectopic bone growth. Many BMPs are part of the transforming growth factor-beta (TGFB) superfamily. BMPs were originally identified by an ability of demineralized bone extract to induce endochondral osteogenesis in vivo in an extraskeletal site. Based on its expression early in embryogenesis, the BMP encoded by this gene has a proposed role in early development. In addition, the fact that this BMP is closely related to BMP5 and BMP7 has lead to speculation of possible bone inductive activity. -

What is bmp6 protein?
Function: BMP6 stimulates bone formation. It signals through BMP receptors and Smad proteins to induce the expression of osteoblast genes and promote the differentiation of stem cells into osteoblasts (bone-forming cells).
Sources: BMP6 is produced primarily in the liver and circulates in the blood. It also acts locally in bones and other tissues.
Role in bone homeostasis: BMP6 helps maintain bone mass by stimulating bone formation. It counters the bone resorption action of osteoclasts (bone-breaking cells).
Clinical significance: Mutations or deficiencies in BMP6 are associated with reduced bone mineral density and increased fracture risk. BMP6 treatment can help repair bone injuries.
What is the function of bmp6 protein?
Bone formation: BMP6 stimulates stem cell differentiation into osteoblasts, the bone-forming cells. This increases bone matrix production and mineralization.
Bone homeostasis: It counters bone resorption by osteoclasts, maintaining balanced bone remodeling and homeostasis.
Osteoblast regulation: BMP6 induces the expression of genes involved in osteoblast differentiation and function, like Runx2, osterix, osteocalcin etc.
Iron metabolism: It acts as a key regulator of systemic iron balance by inducing hepatic hepcidin expression, which reduces intestinal iron absorption.
Bmp6 related signaling pathway
BMP pathway: BMP6 binds to type 1 (ALK2/3/6) and type 2 (BMPR2) serine/threonine kinase receptors.
Smad pathway: Receptor activation phosphorylates regulatory Smads (R-Smads) 1, 5, and 8. Phospho-R-Smads complex with Co-Smad4 and translocate to the nucleus.
Target gene expression: The Smad complex associates with transcription factors like Runx2 to induce genes responsible for osteoblast differentiation (ALP, OC, OPG etc).
Wnt pathway: BMP6 signaling intersects with the Wnt/β-catenin pathway important for osteogenesis and bone homeostasis.
